The fluorogenic peptide substrate DABCYL-Abu-HPFHLVIHT-EDANS has been developed to continuously measure the proteolytic activity of human renin (Km = 1.5 μM at physiological pH). Cleavage of the FRET substrate occurs specifically at the Leu-Val bond and corresponds to the renin cleavage site of angiotensinogen. By means of this assay as low as 30 ng/mL renin can be detected after an incubation of only 3-5 min. Enzymatic cleavage yields the EDANS-fragment M-2645.
